Abstract:
Surgeons must rely on their naked eye and experience during often-lengthy procedures to avoid sensitive glands and tissues. Also, they must depend on biopsy results to determine if the diseased tissue has been removed. Molecular modalities imaging such as SPECT/PET, MRI, and ultrasound give good preoperative images, but cannot be used intraoperatively. Optical imaging of NIR region requires highly selective fluorophores which can target specific tissues. Fluorophores in the near infrared window offer improved tissue penetration. The development of fluorophores that can target diseased and healthy tissues would allow clear delineation. These contrast agents have great potential to be used in biomedical applications including cancer.
Through manipulations of the cyanine scaffold, we have synthesized zwitterionic cyanine imaging agents with low background fluorescence for potential use during surgery. The zwitterionic fluorophores allow for rapid renal clearance and a site of modification to be conjugated to tumor targeting ligand such as CRGD or antibodies. Using tumor targeting ligands, there is high accumulation of fluorescent dye in the diseased cells for the surgeon to remove the tumor tissues from the healthy ones.
